FDA Warning Letter: API Process Validation, Batch Records, Analytical Methods, and Stability Deficiencies

What happened

FDA states that its records review identified significant CGMP deviations at Tianjin Kilo Pharmaceutical Sci-tech Co., Ltd., an API manufacturer. The firm manufactured and distributed APIs to U.S. 503A compounding pharmacies without adequate process validation; it had described the APIs as still in development and said formal documents, including SOPs, were not finalized. FDA also found deficient production documentation: one batch record omitted critical processing information, including redacted details and major equipment, while another requested batch record was not provided. In addition, the firm had not validated, or where applicable verified, each analytical method used for drugs distributed to the United States. FDA said this undermined assurance that customer data accurately reflected identity, strength, quality, purity, potency, and safety. The firm also lacked routine stability testing and did not provide stability data for a redacted API, leaving insufficient evidence that APIs retained specifications through labeled expiry. FDA requested validation remediation, detailed master batch records, an independent laboratory assessment, and a stability CAPA plan. FDA placed all drugs offered for U.S. import from the firm on Import Alert 66-40 on July 21, 2026.

Why it matters

From a GMP perspective, the cited gaps affect the evidence needed to show that API manufacturing remains controlled and reproducible. Missing or incomplete batch records can hinder monitoring of intra-batch and inter-batch variation and reconstruction of significant manufacturing steps. Unvalidated or unverified analytical methods can weaken confidence in release and quality data, while absent routine stability studies leave shelf-life and storage claims without adequate scientific support. Together, these issues may limit assurance that distributed APIs consistently meet predetermined quality attributes and established specifications.
